Prevention of skeletal-related events (pathological fractures, spinal compression, radiation or surgery to bone, or tumor-induced hypercalcemia) in patients with advanced malignancies involving bone. ZOMETA is indicated for multiple myeloma and bone metastases in more than 60 countries, including the United States and the European Union.

ZOMETA® is a highly potent bisphosphonate, (biss-foss-fon-ate). Bisphosphonates are a class of drugs that slow the bone-destroying activity that occurs with bone metastases. They directly work against the abnormal cells that cause the "wearing away" (resorption) of bone and help slow down the abnormal build-up of unstable bone. Bisphosphonates are used to help improve bone strength in many diseases associated with bone resorption, including cancer.

ZOMETA powder for solution for infusion is contraindicated in pregnancy, breast-feeding women, in patients with clinically significant hypersensitivity to zoledronic acid, other bisphosphonates or any of the excipients in the formulation of ZOMETA.

ZOMETA® must only be used by clinicians experienced in the administration of intravenous bisphosphonates. The recommended dose in the prevention of skeletal related events in patients with advanced malignancies involving bone is 4 mg reconstituted and further diluted ZOMETA solution for infusion (diluted with 100 ml 0.9 % w/v sodium chloride or 5 % w/v glucose solution), given in no less than a 15-minute intravenous infusion every 3 to 4 weeks.

Frequencies of adverse reactions for ZOMETA 4 mg are mainly based on data collection from chronic treatment. Adverse reactions to ZOMETA are similar to those reported for other bisphosphonates and can be expected to occur in approximately one third of patients. Intravenous administration has been most commonly associated with a flu-like syndrome in about 9 % of patients, including bone pain (9.1 %), fever (7.2 %), fatigue (4.1 %) and rigors (2.8 %).

ZOMETA (zoledronic acid) injection is indicated for the treatment of patients with multiple myeloma and patients with documented bone metastases from solid tumors, in conjunction with standard antineoplastic therapy. Prostate cancer should have progressed after treatment with at least one hormonal therapy. ZOMETA (zoledronic acid) injection is indicated for the treatment of hypercalcemia of malignancy.

ZOMETA is given by intravenous infusion. It can be administered in your doctor's office, a clinic, or hospital as outpatient therapy. The infusion takes at least 15 minutes. ZOMETA is administered every 3 to 4 weeks. Before ZOMETA was available, IV bisphosphonate therapy took at least 2 hours to administer.

ZOMETA was tested in three large studies involving over 3,000 patients. These patients all had bone metastases from solid tumors (breast, prostate, lung, kidney cancer, genitourinary cancer, bladder cancer, colorectal cancer, other gastrointestinal cancers, liver cancer, head and neck cancer, malignant melanoma, sarcoma, and others) or multiple myeloma. They were all receiving chemotherapy or hormonal therapy.

The most common side effects are flu-like symptoms that are usually mild and transient. These symptoms include fatigue, nausea, vomiting, bone pain, or lack of appetite; they may start after the 1st infusion and can last 3 to 5 days. These flu-like symptoms usually occur with the 1st, and perhaps 2nd dose. Patients who experience these symptoms usually do not experience them with the 3rd or 4th dose. If necessary, your doctor can prescribe a mild pain reliever to make you more comfortable.

Tarceva monotherapy is indicated for the treatment of patients with locally advanced or metastatic Non-Small Cell Lung Cancer (NSCLC) after failure of at least one prior chemotherapy regimen. Results from two multicenter, placebo-controlled, randomized, Phase III trials conducted in first-line patients with locally advanced or metastatic NSCLC showed no clinical benefit with the concurrent administration of Tarceva with platinum-based chemotherapy, and its use is not recommended in that setting.
