For a printer to work on your Mac, your Mac's operating system needs to have a "driver" for that printer. Additional software may be needed as well: check your operating system manual for details. Yes! Generally you'll need at least Mac OS X. There's a special version for Mac (the BS*-mac.sitx) which bandles all necessary files into a Mac application and should be recognized by the OS. You can't print directly to it through the Chooser from a Mac. You must print to the software RIP, which is running on an iMac next to the printer. The correct spool is "DesignJet 500PS 42@junco" in the Biology zone of the Chooser. The correct PPD for the printer is HP500P42.PPD. You can find this on Squirrel Public. The Mac OS X installer is asking for a username and password for an account on that computer that has administrator access. This proves that you have permission to install the software on that computer. This account is usually the account you use to log into that computer. If your computer is set up to log into an account automatically, you will need to know that password.

Native printer drivers are not supported. At one time Wine supported 16bit native drivers but that was long ago. Wine uses the printers (and other devices) installed in your operating system. For the most part if you don't have the device installed on your OS then wine can't use it.

The very short answer is: you can't do this easily. There are problems between the USB-to-serial stuff; problems with printer drivers; and problems with OS X (or OS 9, etc) not knowing about either.
