If you are programming for MS-DOS, you will need to write an interrupt driven handler for the RS-232 ports. Only the most trival output-only applications can use the BIOS functions built into the PC. Anything that needs reliable input needs to use interrupts. Win16 and Win32 supply drivers that take care of that problem. The serial port is a 9-pin D shaped connector, similar to the video connector except with less pins. Usually, there are two on a PC. One may possibly be in use by the mouse (mainly on older machines). They are also referred to as Com ports or Communications ports. The 10-pin header next to the dip switches is a 2nd serial port. You can hook this up to your PC's serial port (with a null modem), and get connectivity that way, utilizing the 'qtalk' app on the i-opener. This seems to support zmodem, but the i-opener doesn't seem to have the sz/rz binaries. If it did (or if you can find/FTP them), it might make an easy way to transfer files to/from your PC.
