It depends on a couple of things, the size of the balloons and the tank size. Temperature and tank pressures are also factors. The chart below will give you an approximate number of balloons based on these factors. The chart assumes that the tank is full and that you're not over-inflating the balloons. Note: You won't get 125 balloons if the tank is half full and the 9'' balloons you've bought are inflated to twice that size.

Helium inflated balloons in a helium environment will last as long as air filled balloons last in an air environment. Generally round latex balloons last as long as the Sparklet lasts (3 - 5 weeks). Hearts last for 3 weeks (inflate them nice and full to get a full "heart" shape). 350's last for 2 weeks and 260's last for 1 week.

You must inflate 100% wrinkle-free using the same equipment you use to inflate your latex balloons. NEVER use the automatic shutoff inflator for the OUTER Bubble because it does not have enough power to inflate a Bubble 100% wrinkle-free. The automatic shutoff should only be used for the INNER balloon.

Can I inflate the latex balloons with air? Will this lessen the float time?

If you inflate the inserted latex balloons with air, AND the Sparklet with helium, the inner latex balloons will grow until they finally POP (usually within the first day). You MUST inflate both the inserted latex and outer Sparklet with helium. If you do not need the Sparklet to float, you can use nitrogen (or compressed air) in BOTH the inner latex and outer Sparklet. Always use the same gas in both inner and outer balloons.
